If I applied a couple of layers of TR 104 High Temp mold release over a gelcoat part and then applied a couple of layers of MAT with polyester resin, will I be able to release the fiberglass cloth leaving a clear finish on the gelcoat?

I want the original gelcoat surface not to be damaged and get a male mold out of it.

Personally I like a good old car paste wax. Tried some spray-on mold release one time and found parts hard to separate. I went back to the old can of Turtle wax I had laying around the garage.

Yes, is the answer to your question. There are mold release waxes designed for this type of work and also PVA, which is a sprayable polyvinylalcohol solution used for mold releasing. I've made many a parts from original boat pieces.

If I recall correctly, the instructions on the item I was considering mentioned buffing, perhaps after 2nd & 3rd application for a total of 4.
